Expression of high mobility group protein box 1 at different time after cerebral ischemia-reperfusion

Abstract:

Objective

To study the expression of high mobility group protein box 1 (HMGB1) at different times in cerebral ischemia-reperfusion model of rats.

Methods

Forty male SD rats were divided into 6 h group, 24 h group, 3 d group and 7 d group by random number table method. The rat model of focal cerebral ischemia-reperfusion injury was established by occlusion of the middle cerebral artery with thread occlusion for 2 h. The volume of cerebral infarction was measured by TTC staining. The mRNA level of HMGB1 in rat brain tissue was measured by semi-quantitative RT-PCR, and the protein expression of HMGB1 was detected by ELISA and Western blotting at different times after reperfusion in rat middle cerebral artery acclusion (MCAO) model.

Results

The mRNA level and protein level of HMGB1 decreased at 6 h-7 d after ischemia-reperfusion. The mRNA level of HMGB1 in the right brain tissue was higher than that in the left brain tissue at 6 h and 24 h after cerebral ischemia reperfusion. The protein level of HMGB1 in the right brain tissue was higher than that in the left brain tissue 6 h after cerebral ischemia reperfusion.

Conclusion

The level of HMGB1 decreased at 6 h-7 d after cerebral ischemia-reperfusion.
